Ovarian and Adrenal Androgen Biosynthesis and Metabolism

چکیده
The pathways of adrenal and ovarian steroid biosynthesis use the same enzymes for the initial steps of steroidogenesis but express different enzymes that convert steroid precursors to the final active products. Both the adrenal and ovary produce dehydroepiandrosterone (DHEA), the principal precursor of androgens and estrogens. The key enzyme in DHEA production is P450c17, which catalyzes both 17α-hydroxylation and 17,20lyase activities. The 17,20-lyase activity of human P450c17 strongly favors 17-hydroxypregnenolone rather than 17-hydroxyprogesterone (17-OHP) as a substrate, producing abundant DHEA, so that most human androgens and estrogens derive from DHEA. Understanding the biochemistry of P450c17 is central to understanding the hyperandrogenism of polycystic ovary syndrome (PCOS). Rare genetic disorders of steroidogenesis provide human genetic knockout experiments of nature, yielding important information about the biosynthesis and physiological roles of steroids.

Age-related Androgen Secretion in Healthy Women and in Women with Polycystic Ovary Syndrome
The number of ovarian follicles declines with age resulting in a significant decrease of fertility by the age of 40. However, the age when follicle loss starts to affect ovarian endocrine function is not well recognized. The purpose of the present study was to investigate age-related ovarian/adrenal androgen secretion, which is crucial for estrogen biosynthesis in healthy women and in women wit...
متن کاملThe role of androgen therapy.
The concept of a female androgen insufficiency syndrome, although not new, remains somewhat controversial. Androgens are quantitatively the predominant sex steroid in women, circulating in the micromolar and nanomolar concentration range, compared with picomolar levels of oestrogens. The most significant biologically active androgen is testosterone (T), which circulates bound tightly to sex-hor...
متن کاملP-216: Common Polymorphisms of StAR Gene Are Not Associated with Polycystic Ovary Syndrome in Iranian Patients
Background: Polycystic ovary syndrome (PCOS) is one of the most common endocrine disorders, affecting 5-10% of reproductive aged women. High levels of androgen hormone can be accounted as the most important symptoms in PCOS. Androgen is the precursor of steroid biosynthesis in ovary. The transport of cholesterol to the inner mitochondrial membrane through StAR protein is also necessary for andr...

OBJECTIVE To describe the sources, production rates, circulating concentrations, and regulatory mechanisms of the major androgen precursors and androgens in women. DESIGN Review of the major published literature. RESULT(S) Quantitatively, women secrete greater amounts of androgen than of estrogen. The major circulating steroids generally classified as androgens include dehydroepiandrosteron...
متن کاملBeyond adrenal and ovarian androgen generation: Increased peripheral 5 alpha-reductase activity in women with polycystic ovary syndrome.
Hyperandrogenism, a main clinical feature of polycystic ovary syndrome (PCOS), is thought to result from enhanced ovarian and adrenal androgen generation. To investigate the contribution of peripheral steroidogenesis, we used an oral challenge with dehydroepiandrosterone (DHEA) and analyzed its downstream conversion toward androgens in eight women with PCOS (age, 20-32 yr; body mass index, 20-4...
